Interested parties can contact assistant core director to discuss preliminary approach of intended creation of a mouse model. Primary requirement to generate a mouse model at BCH using mouse core services is the PI must have an approved IACUC and IBC protocol for the desired model. After approval is granted the mouse core will co-ordinate with PI or lab member to submit request form and provide required reagents before scheduling an experiment.

Requirements

  1. Approved IACUC and IBC protocol for the desired mouse model.
  2. Submit electronic request form (eForm) for services signed by PI and designated lab member.
  3. Mouse core will co-ordinate with PI or lab member to collect required reagents before scheduling an experiment.
  4. A fund number or purchase order number to reimburse fees for requested services.

Current Services

Core provide rederivation services in following categories-

  1. Imported frozen embryos
  2. Imported frozen sperm
  3. Imported live mouse strain (Quarantined)
  4. In-house frozen embryos
  5. In-house frozen sperm

Frozen embryos at 2-cell to 8-cell stage thawed and transferred to specific pathogen free (SPF) recipient females and housed in an isolated room. Tail snip biopsies from pups provided at P7 to confirm genotype. After pups are weaned the recipient female is tested for pathogen by ARCH Veterinarian technicians. Test results evaluated by ARCH and approved pathogen free colony is released to the investigator for further propagation.

In vitro fertilization:

Imported or in-house frozen sperm is fertilized in vitro using eggs from pathogen free donor females. Fertilized eggs are allowed to incubate overnight in CO2 incubator and following morning scored for 2-cell stage embryos. The embryos are transferred to pathogen free recipient females that are housed in an isolated room. Tail snip biopsies provided at P7 to confirm genotype. After pups are weaned the recipient female is tested for pathogen by ARCH Veterinarian technicians. Test results evaluated by ARCH and approved pathogen free colony is released to the investigator for further propagation.

Mycoplasma sp. Test:

Mouse core strictly follow ARCH Guidance and only accept cell lines that are free of Mycoplasma sp. We recommend sending cell lines to a commercial vendor to test and certify absence of Mycoplasma sp in the cell lines to be used at the Mouse Gene Manipulation Core. We will not accept any cell line without appropriate documentation. Users may contact following vendor or other vendors for test details:
